Bovine collagen type VI, isolated from bovine placenta by pepsin treatment and extraction into dilute acetic acid followed by differential salt precipitation, supplied lyophilised and salt-free at >90% purity, 1 mg. Type VI is the beaded-filament collagen of the pericellular matrix, forming a microfibrillar network that anchors cells — chondrocytes, myofibres, adipocytes — to the surrounding interstitial matrix and binds collagen types I, II and IV, fibronectin, biglycan and decorin. Use as an ELISA / Western blot standard, an immunoprecipitation antigen or a cell-culture coating substrate. STORAGE / RECONSTITUTION
Collagen type VI dissolves very slowly in 0.5 M acetic acid and requires both vigorous mixing and time. Add a small volume of 0.5 M acetic acid, pH 2.5, and use a Potter homogeniser (or similar) to break the protein cake into suspension. Add further 0.5 M acetic acid, pH 2.5, to a concentration of 0.1 mg/mL and stir vigorously for several days. Concentrations above 0.1 mg/mL require even longer; use 6 M urea (or another chaotropic agent) if the application allows.
Lyophilised protein: ships at ambient temperature; store long-term at −20 °C or lower (stable for 2 years).
Reconstituted protein: store at +4 °C for up to 4 weeks, or aliquot and freeze.
Avoid repeated freeze-thaw cycles of reconstituted material.
