Purification
Collagen was extracted
from washed dissected tissue into dilute acetic acid after pepsin
treatment. Collagen type VI was purified by using differential salt
precipitation

Reconstitution
Collagen type VI dissolves
very slowly in 0.5 M acetic acid and requires both vigorous mixing
and time.
Add small amount of 0.5 M
acetic acid, pH 2.5 and use Potter homogenizer (or similar) to break
up the protein cake into suspension. Add 0.5 M acetic acid, pH 2.5 to
yield 0.1 mg/ml concentration and use vigorous stirring for several
days.
Concentration higher than
0.1 mg/ml in acetic acid requires even longer time. Use 6M urea (or
other chaotropic agent) if application allows it.
Storage
Lyophilized protein:
shipping at ambient temperature, long term storage (2 years) at -20ºC
or lower. Storage of reconstituted protein 1 month at +4ºC
