Rabbit polyclonal antibody against native human collagen type I + III (heterotypic blend, COL1A1 + COL3A1), affinity-purified on immobilised human collagen I+III and cross-absorbed against human collagen II, IV, V and plasma proteins, 0.05 mg. Dual-specificity single-pool reagent for the native I/III heterotypic-fibril readout — dermal fibrosis, scar maturation, vascular adventitia, intestinal and organ fibrosis, and tumour stroma. For sandwich ELISA, pair with cat. 478 (mouse pan-fibrillar mAb capture) or the isotype-specific cat. 380 / cat. 393 mAbs. Validated for ELISA (1:4 000), frozen-section IHC (1:30) and ECM immunostaining in cell culture — not suitable for denaturing conditions.
SPECIFICATIONS
Catalogue number: 198 | Host: Rabbit | Target: Human
Target antigen: Native human collagen type I (COL1A1) + type III (COL3A1), triple-helical fold
Gene: COL1A1 (17q21.33), COL3A1 (2q32.2) | UniProt: P02452, P02461
Clonality: Polyclonal — dual-specificity (balanced anti-I + anti-III titres) | Conjugate: Unconjugated
Specificity: Binds human collagen I + III; type IV cross-reactivity < 3 %, type V < 2 %, type II cross-absorbed, no reactivity to plasma proteins. Native-conformation — not suitable for denaturing / dissociating conditions.
Purification: Affinity-purified on immobilised human collagen I + III, cross-absorbed against collagen II, IV, V and plasma proteins.
Format: Lyophilised from 0.01 M Na phosphate, 0.01 M Na borate, 0.15 M NaCl, 1% dextran, 1% mannitol, pH 8 | Package size: 0.05 mg (0.1 mg/mL after reconstitution in 0.5 mL)
STORAGE / RECONSTITUTION
Reconstitute with 0.5 mL sterile pure water (0.1 mg/mL working stock); mix gently by inversion, do not vortex. Ships at ambient temperature. Store lyophilised antibody 2 years at -20 °C or lower; store reconstituted antibody at +4 °C up to 4 weeks, or aliquot and freeze at -20 °C. Avoid repeated freeze-thaw.
APPLICATIONS
- ELISA — detection of human collagen I + III (starting dilution 1:4 000); a single pool detects both fibrillar collagens at balanced titre. For sandwich ELISA, pair with cat. 478 (pan-fibrillar mAb capture) or isotype-specific cat. 380 / cat. 393 mAbs.
- IHC (frozen sections) — human skin, dermal scar, vascular adventitia, intestinal wall, granulation tissue, lung parenchyma and tumour stroma (starting dilution 1:30); single-stain heterotypic-fibril detection, no antigen retrieval typically required.
- Cell culture / ECM staining — heterotypic I/III networks secreted by fibroblasts, hepatic stellate cells and vascular smooth-muscle cells; single-channel reporter of fibrillar-collagen matrix deposition.
- Note: native-conformation antibody — not validated for FFPE or denatured Western blot.
APPLICATION HIGHLIGHTS — HIGH-VALUE RESEARCH NICHES
- Dermal fibrosis / scar maturation — the I/III ratio is a key quantitative readout of scar maturation.
- Vascular adventitia and aneurysm research — principal site of heterotypic I/III collagen in the arterial wall.
- Intestinal / IBD fibrosis.
- Hepatic / pulmonary / renal fibrosis.
- Tumour stroma.
- ECM deposition by primary cultured fibroblasts.
- Dupuytren, adhesive capsulitis, keloid.
