ELISA capture antibody (mouse total-Ig)
• Coat at 1–5 µg/mL in carbonate / bicarbonate coating buffer pH 9.6.
• For detection-arm ELISA, dilute 1:30 000 in blocking / sample buffer.
• Validated in hybridoma-supernatant screening and mouse-serum titration.
ELISA detection (in combination with HRP-conjugated secondary)
• 1:30 000 dilution as above; use a downstream anti-goat HRP for detection
(YO cat. 319 — rabbit anti-goat IgG, IgM, IgA HRP, 1.2 mg).
Western blot
• 1:1 000 to 1:5 000 dilution in TBS-T + 5 % non-fat milk or 5 % BSA.
• Use as the detection arm against blotted mouse mAb, blotted mouse plasma/serum samples, or hybridoma-secreted mouse Ig.
Immunohistochemistry / Immunofluorescence
• Titrate locally; typical starting dilution 1:200 to 1:1 000.
