APPLICATIONS AND RECOMMENDED WORKING DILUTIONS
ELISA capture — human total-Ig
• Coat at 1–5 µg/mL in carbonate / bicarbonate coating buffer pH 9.6.
• The natural application for the unconjugated pan-Ig polyclonal: total-IgG + IgM + IgA capture from serum / plasma / cell-conditioned media, with isotype-specific HRP secondary antibodies on the detection arm to read out class distribution.
• Validated workflow for vaccine-response total-Ig titration, infectious-disease serology assay development, autoimmune-disease panel construction, and IVIG quality-control.
ELISA detection
• 1:5 000 to 1:30 000 dilution in blocking / sample buffer.
• Use as detection arm against plate-bound human Ig, with a downstream anti-rabbit HRP secondary for chromogenic / chemiluminescent readout.
Western blot
• 1:1 000 to 1:5 000 dilution in TBS-T + 5 % non-fat milk or 5 % BSA.
• Detection of human IgG heavy chain (~50 kDa) and light chain (~25 kDa), IgM µ chain (~70 kDa), and IgA a chain (~55 kDa) under reducing conditions.
Immunohistochemistry / Immunofluorescence
• Starting dilution 1:200 to 1:1 000; titrate locally.
• Plasma-cell IHC marker in lymph-node biopsies, bone-marrow biopsies, plasmacytoma, multiple-myeloma diagnostic panels, and lymphoplasmacytic-lymphoma characterisation.
• Tissue-bound immune-complex detection in nephritis (immune-complex glomerulonephritis), vasculitis, and autoimmune dermatoses (direct immunofluorescence on skin biopsies).
Immunoprecipitation
• The unconjugated affinity-purified format is preferred over HRP / biotin conjugates for IP — couple to Protein A or Protein G beads.
Custom conjugation starting material
• The 50 % glycerol, sodium-phosphate / NaCl liquid format is compatible with standard NHS-ester (HRP, biotin), maleimide (cysteine-thiol), and amine-directed Alexa Fluor / DyLight conjugation chemistries after a buffer-exchange step into the kit-specified reaction buffer.
Sandwich-ELISA detection / capture pairings
• For total-human-Ig sandwich ELISA development, cat. 172 (rabbit) and cat. 133 (goat anti-mouse pan-Ig) cover the most common host-pairing needs. Combine with isotype-specific YO anti-human IgM / IgG / IgA detection arms for class profiling.
APPLICATION HIGHLIGHTS — HIGH-VALUE RESEARCH NICHES
- Infectious-disease serology assay development. Pan-class capture lets a single coating reagent serve as the base for IgG / IgM / IgA isotype-specific detection of anti-pathogen antibodies — the dominant assay architecture for COVID-19, dengue, EBV, CMV, HIV, syphilis, Lyme, and tropical-disease serology kits.
- Autoimmune-disease total-Ig panels. Single-reagent capture for ANA, anti-dsDNA, anti-CCP, anti-Sm, anti-Ro/La, anti-thyroid-peroxidase, anti-transglutaminase isotype-class profiling.
- Vaccine-response titre quantification. Total-IgG / IgM / IgA capture for primary, booster, and durability vaccine-immunogenicity readouts.
- IVIG quality control. Pan-Ig capture as the reference detection chain in IVIG / SCIG batch-release immunoassays.
- Multiple-myeloma / monoclonal-gammopathy IHC. Pan-class staining of plasma cells in bone-marrow biopsies, lymph-node biopsies, plasmacytoma, MGUS, smouldering myeloma, and lymphoplasmacytic lymphoma.
- Immune-complex disease direct-IF. Skin-biopsy direct immunofluorescence for pemphigus, bullous pemphigoid, dermatitis herpetiformis, and lupus erythematosus; kidney-biopsy direct-IF for immune-complex glomerulonephritis, lupus nephritis, IgA nephropathy, and post-infectious glomerulonephritis.
- B-cell biology research. Surface- and secreted-Ig quantification in B-cell development, germinal-centre biology, and B-cell-malignancy phenotyping (chronic lymphocytic leukaemia, follicular lymphoma, marginal-zone lymphoma).
STORAGE
Shipped at ambient temperature.
Liquid antibody (1 mg/mL in PBS + 50 % glycerol): store up to 2 years at -20 °C or lower.
The 50 % glycerol formulation does not freeze solid at -20 °C — no freeze–thaw concerns for occasional aliquot withdrawal.
For long-term archiving and assay-batch consistency, prepare single-use aliquots and store at -80 °C.
