Rabbit polyclonal antibody against human urokinase-type plasminogen activator (uPA / PLAU; abbokinase), affinity-purified on immobilised human urokinase and cross-adsorbed against plasma proteins, 0.5 mg. Pan-form neutralising reagent — recognises HMW single-chain (54 kDa), HMW two-chain (52 kDa) and LMW (33 kDa) uPA and completely inhibits uPA-catalysed plasminogen activation and S2444 cleavage, with no cross-reactivity to tPA, plasmin, trypsin, chymotrypsin, elastase or thrombin. For uPA ELISA, IHC, Western blot, immunoprecipitation and function-blocking studies — the polyclonal companion to the uPA monoclonals cat. 335 (clone PGM2005) and cat. 381 (clone PGM2001).

STORAGE / RECONSTITUTION
Reconstitute with 1 mL sterile pure water. Ships at ambient temperature. Store lyophilised antibody 2 years at -20 °C or lower; store reconstituted antibody at +4 °C up to 4 weeks, or aliquot and freeze at -20 °C. Avoid repeated freeze-thaw.
APPLICATIONS
- ELISA — detection of human uPA (dilution 1:5 000).
- IHC (FFPE / frozen sections) — uPA in tumour invasive fronts and peri-tumoural stroma (breast, colorectal and other carcinomas); starting dilution 1:50.
- Western blot — 1:1 000 to 1:5 000; detects HMW-scuPA (54 kDa), HMW-tcuPA (52 kDa) and LMW-uPA (33 kDa).
- Immunoprecipitation — protein A / G beads, 1–5 µg antibody per 100–500 µL sample (plasma, urine, conditioned medium).
- Functional neutralisation / function-blocking — titrate in molar excess vs uPA; blocks uPA-catalysed plasminogen activation in cancer-cell invasion (transwell, Matrigel, 3-D spheroid) and ECM-degradation assays.
