Collagenase from king crab (Paralithodes camtschatica) hepatopancreas, lyophilised, 1 mg — a mixture of cold-adapted proteolytic enzymes for gentle dissociation of animal cells from tissue and from cultureware. The marine, cold-active enzymes work efficiently at low temperature (from 4 °C) and over a broad pH range, allowing tissue dissociation under conditions that better preserve surface antigens and viability than warm bacterial-collagenase digests, and they are readily and completely inhibited by DFP, PMSF or EDTA when digestion must be stopped.
SPECIFICATIONS
Catalogue number: 206 | Source: King crab (Paralithodes camtschatica) hepatopancreas
Composition: Mixture of proteolytic (collagenolytic) enzymes
Purity: Five bands in the range 18–34 kDa
Purification: Acetone extraction; affinity and ion-exchange chromatography

STORAGE / RECONSTITUTION
Dissolve in buffer containing 1–10 mM CaCl2 at pH 6–9. Ships at ambient temperature. Store lyophilised enzyme 2 years at −20 °C or lower. Avoid repeated freeze-thaw of the reconstituted enzyme.
APPLICATIONS
- Isolation of primary cells from tissue — gentle enzymatic dissociation at 4–40 °C.
- Detachment of adherent cells from cultureware, including from collagen-coated surfaces and 3D collagen gels.
- Release of cells and spheroids from collagen hydrogels, e.g. gels cast from cat. 130 or cat. 176.
- Controlled digestion of collagen substrates in matrix-degradation and ECM-turnover experiments.
- Sample preparation for flow cytometry where preservation of surface epitopes matters — low-temperature digestion limits epitope loss.
- Stop the reaction with EDTA, PMSF or DFP for defined digestion endpoints.
APPLICATION HIGHLIGHTS — HIGH-VALUE RESEARCH NICHES
- Primary-cell isolation — cold-active digestion for fragile primary and stem-cell preparations.
- Immunophenotyping — surface-antigen-preserving dissociation ahead of flow cytometry and single-cell sequencing.
- 3D culture workflows — recovery of viable cells and organoids from collagen gels and scaffolds.
- Marine-enzyme biotechnology — cold-adapted protease research and process development.
- ECM biology — controlled collagen degradation and matrix-remodelling assays.
